This study evaluated the effects of dose of equine chorionic gonadotropin (eCG) and its splitting in different days of the synchronization protocol on reproductive performance of primiparous and multiparous Nellore cows. In the present study, 2,536 Nellore cows (1,634 primiparous and 902 multiparous) were assigned to receive in a 2 × 2 factorial design 1) an intravaginal progesterone (P4) device and 2.0 mg of estradiol benzoate (EB) on day -11, 12.5 mg (i.m.) of dinoprost tromethamine (PGF), 300 IU (i.m.) of eCG, 0.6 mg (i.m.) of estradiol cypionate (ECP), and P4 device withdrawal on day -2, followed by TAI on day 0 (n = 632 cows, being 409 primiparous and 223 multiparous; 300-2), 2) 300 IU (i.m) of eCG administered on days -4 and -2 (150 IU of eCG/day; n = 637 cows, being 412 primiparous and 225 multiparous; 300-4-2), 3) 400 IU (i.m.) of eCG administered on day -2 (n = 633 cows, being 406 primiparous and 227 multiparous; 400-2), and 4) 400 IU (i.m) of eCG administered on days -4 and -2 (200 IU of eCG/day; n = 634 cows, being 407 primiparous and 227 multiparous; 400-4-2). Individual cow BCS was assessed on days -11, 0 (timed-AI), and 31 of the study. Body condition score of the animals was classified into LOW or HIGH using the threshold of 2.75 (≤2.75 = LOW; >2.75 = HIGH). For primiparous cows, an eCG splitting effect was observed on follicle size, as cows receiving eCG on days -4 and -2 of the synchronization protocol had a larger follicle than cows administered eCG only on day -2. For day 31 P/AI, primiparous cows receiving 400-4-2, regardless of BCS, had a greater P/AI than cows from other treatments. Administering 400-4-2 to LOW BCS cows also resulted in greater P/AI than all other treatments assigned to LOW BCS cows. For multiparous cows, no treatment effect was observed for follicle size, estrus expression, and day 31 P/AI (P ≥ 0.21). In summary, increasing the dose and splitting the dose of eCG positively impacted the pregnancy rates of primiparous cows under a BCS ≤2.75, but no effects were detected on multiparous cows.

Three experiments were designed to evaluate the effects of conventional and sexed-semen on reproductive performance of dairy females detected in estrus. In Exp. 1, 978 lactating Holstein cows and 116 Holstein heifers were used. Cows or heifers were randomly assigned to receive conventional (CONV) or sexed (SEXD) semen on day 0 of a TAI protocol. The TAI was performed using commercial CONV or SEXD frozen-thawed semen from the same 6 Holstein bulls, and SEXD semen was sexed through fluorescence cytometry. In Exp. 2, 390 lactating Holstein × Gir dairy cows and 344 Holstein × Gir heifers were used. Cows and heifers were randomly assigned to receive CONV or SEXD semen on day 0 with frozen-thawed semen from 5 Holstein × Gir bulls. The SEXD was sorted through the Sexcel™ methodology. In Exp. 3, 789 primiparous lactating Holstein cows were enrolled. Cows were randomly assigned to receive CONV or SEXD semen from 5 Holstein bulls, and the SEXD semen was sexed using the same semen allocation methodology described in Exp. 2. All data were analyzed using SAS. Regardless of the experiment, no treatment × bull interaction was observed for any of the variables analyzed herein (P ≥ 0.20). In Exp. 1, multiparous cows assigned to CONV had a greater P/AI vs. cohorts assigned to SEXD (P < 0.01) and a similar response tended to be observed for secundiparous cows (P = 0.10). No treatment effects were further observed when the effects of semen on day 60 P/AI were stratified by heat stress, milk yield, and CL at the beginning of the protocol. In Exp. 2, no P/AI differences between treatment were detected either on days 32 (P = 0.32) or 60 of pregnancy (P = 0.20). In Exp. 3, cows assigned to SEXD had reduced P/AI on day 32 (P = 0.03) and tended to have a reduced P/AI on day 60 of pregnancy (P = 0.06). No further treatment effects or interactions were observed herein (P ≥ 0.13). Overall P/AI of SEXD semen was 78% of the CONV (P < 0.01), but the variation of these results requires some attention. Moreover, an additional analysis was performed considering the median value of P/AI in bulls assigned to CONV and SEXD, and bulls that had a greater P/AI in CONV also had a greater P/AI in SEXD (P = 0.03). In summary, although not directly comparable, the sexing technologies herein provided similar results in heifers, but not in other categories (primiparous, secundiparous, and multiparous cows). Additionally, the P/AI of a sexed semen is associated to its performance on the conventional semen counterpart. Moreover, in the present experiment, no other parameters were shown to impact fertility of dairy females assigned to conventional or sexed-semen.

This experiment was designed to evaluate the effects of increasing doses of estradiol cypionate (ECP) and different body condition score (BCS) on reproductive performance of Bos indicus beef females assigned to a timed-artificial insemination (TAI) management. In this experiment, 1683 Bos indicus Nellore cows were blocked by parity and assigned to receive 1) an intravaginal P4 device (1.9 g of P4) and 2.0 mg of estradiol benzoate on day -11, 12.5 mg (i.m.) of dinoprost tromethamine, 300 IU (i.m.) of equine chorionic gonadotrophin, 0.6 mg (i.m.) of estradiol cypionate and CIDR withdrawal on day -2, followed by TAI on day 0 (n = 849; 0.6ECP) or 2) the same synchronization protocol with 1.0 mg of ECP on day -2 (n = 834; 1.0ECP). In both treatments, estrus expression was measured between days -2 and 0. Body condition score (BCS) was evaluated on days -11, 31, and 71 of the experiment and the BCS variation (Δ) was also determined between these timepoints. Transrectal ultrasonography was performed on days 31, 71, and 111 for pregnancy rate determination. All binary data were analyzed using cow as the experimental unit with GLIMMIX, whereas continuous variables were analyzed with the MIXED procedure of SAS. No treatment effects were observed on estrus expression rate. Treatment × BCS interactions were observed for pregnancy rates in all time points (days 31, 71, and 111), as 1.0ECP cows with a LOW BCS also had a greater P/AI than cows assigned to 0.6ECP. In summary, increasing the dose of ECP benefited the reproductive performance of Nellore beef cows with a reduced BCS (≤2.75), whereas no benefits were seen when the BCS was considered adequate (>2.75).

This study was designed to evaluate whether the utilization of a second PGF2α treatment at the end of an estradiol/progesterone (E2/P4)-based protocol with or without GnRH at the beginning of the protocol would improve pregnancy rates of lactating Holstein cows assigned to timed embryo transfer. A total of 501 lactating Holstein cows in 5 farms were enrolled in the experiment. Within farm, cows were blocked by parity and, within block, were assigned randomly to (1) insertion of an intravaginal P4 device (controlled internal drug-releasing device; CIDR) and estradiol benzoate on d -11, PGF2α on d -4, CIDR withdrawal and an injection of estradiol cypionate on d -2, and timed embryo transfer on d 7 (1-PGF; n = 164); (2) the same treatments as 1-PGF, but with PGF2α administered on d -4 and -2 (2-PGF; n = 171); and (3) 2-PGF with the addition of a GnRH treatment on d -11 (GnRH+2-PGF; n = 166). Ovaries were scanned by transrectal ultrasonography on d -11, -4, and 7, and blood samples were collected on d -11, -4, 0, and 7 for P4 determination. Treatment comparisons were performed using contrasts. The proportion of cows with a new corpus luteum on d -4 was greater in GnRH+2-PGF cows. Cows in 1-PGF had a greater P4 concentration on d 0 but lesser P4 on d 7 compared with cows in the other groups. Cows assigned to receive 2-PGF (2-PGF and GnRH+2-PGF) had greater estrus expression, and a greater proportion of cows ovulated to estradiol cypionate. No further contrast effects were observed for follicle diameter, double ovulation rate, pregnancy per embryo transfer (P/ET) on d 32 and 60, or pregnancy loss. As P4 concentration on d -4 increased, P/ET on d 60 tended to increase. Cows with P4 ≥3.66 ng/mL on d -4 had greater P/ET on d 32 and 60 than those with P4 below that threshold. Regardless of treatment, cows with P4 concentration ≥3.66 ng/mL also had a greater pregnancy per synchronized protocol (P/SP) on d 60. Also, a P4 concentration on d -4 (low or high) × follicle diameter (continuous) interaction tendency was observed when evaluating P/ET. Although P/ET did not differ among cows with different follicles sizes with reduced P4 concentration on d -4 (<3.66 ng/mL), it increased in cows with larger follicles exposed to increased P4 concentration (≥3.66 ng/mL). When P4 on d 0 was evaluated, P/ET on d 32 and 60 was greater for cows with low (≤0.09 ng/mL) versus high (>0.21 ng/mL) P4; as P4 concentration on d 0 increased, P/ET linearly decreased. In summary, cows with increased P4 concentrations during growth of the ovulatory follicular wave had improved P/ET. Administering a second PGF2α dose reduced P4 concentration on d 0 and increased ovulatory response to the protocol, but no benefits were observed on P/ET or P/SP.

This experiment evaluated pregnancy rates to fixed-time artificial insemination (FTAI) in Bos indicus beef cows according to their body condition score (BCS) at calving and subsequent change until 30 days after FTAI. Non-pregnant, suckling Nelore cows (n = 593 primiparous, 461 secundiparous, and 893 multiparous) were evaluated for BCS at calving and FTAI, and at 30 days after FTAI when cow pregnancy status was verified. Cow BCS at calving was subtracted from BCS recorded at pregnancy diagnosis, and cows classified as those that lost BCS (L), maintained BCS (M), or gained BCS (G) during this period. Cows that calved with BCS ≥ 5.0 had greater (P < 0.01) BCS throughout the experiment, and greater (P < 0.01) pregnancy rates to FTAI compared with cows that calved with BCS < 5.0 (54.8 vs. 34.2%). Pregnancy rates to FTAI were greater (P < 0.01) for G and M cows compared with L cows (50.0%, 47.5%, and 36.0%, respectively), and similar (P = 0.46) between G and M cows. Moreover, pregnancy rates to FTAI in G cows that calved with BCS < 5.0 were less compared with L (tendency; P = 0.08) and M cows (P < 0.01) that calved with BCS ≥ 5.0 (42.2%, 48.3%, and 58.3%, respectively). In summary, pregnancy rates to FTAI were greater in B. indicus cows that calved with a BCS ≥ 5.0 regardless of parity and post-calving BCS change, and greater in M and G cows within those that calved with BCS < 5.0 or ≥ 5.0.

This experiment was designed to evaluate the effects of increasing doses of estradiol cypionate (ECP) on reproductive function of lactating dairy cows during the summer. In Exp. 1, 643 lactating Holstein cows were blocked by parity and assigned to receive 1) an intravaginal P4 device (1.9 g of P4) and 2.0 mg of estradiol benzoate on day -11, 25 mg (i.m.) of dinoprost tromethamine on day -4, 1.0 mg (i.m.) of estradiol cypionate and CIDR withdrawal on day -2, followed by TAI on day 0 (n = 326; ECP-1) or 2) the same synchronization protocol with 2.0 mg of ECP on day -2 (n = 317; ECP-2). In both treatments, cows were TAI on day 0 of the protocol, and cow rectal temperature was measured on days -2, 0, and 7. In Exp. 2, 608 lactating crossbred Holstein × Gir dairy cows were blocked by parity and enrolled to the same treatments as in Exp. 1, but on day 7, cows received one viable embryo into the uterine horn. In Exp. 1, a greater percentage of ECP-2 cows were detected on estrus (81.3 vs. 91.1%, respectively). A treatment × body condition score (BCS) interaction was observed on day 60 pregnancy per AI (P/AI), as ECP-2 cows with a BCS <2.75 had a greater P/AI vs. ECP-1, but an opposite result was observed in cows with a BCS ≥2.75. Regardless of treatment, there were effects of mean rectal temperature and heat stress events on P/AI. Treatment affected the diameter of the ovulatory follicle at TAI (ECP-1 = 15.3 mm vs. ECP-2 = 14.8 mm) and a greater percentage of ECP-1 cows had larger follicles (≥16.5 mm), but ECP-2 resulted in a greater incidence of early ovulatory cows (ovulating before day 0). Therefore, follicle diameter at TAI affected P/AI on day 60 in cows receiving ECP-2 and tended to affect ECP-1 cows. A treatment effect was observed on time to estrus following ECP treatments and a reduced proportion of ECP-2 cows showed estrus at TAI. Regardless of treatment, cows detected on estrus 48 h after ECP administration had a greater P/AI on day 60 vs. cows detected on estrus 24 h. In Exp. 2, a greater percentage of ECP-2 cows were detected on estrus (68.3 vs. 81.4%, respectively). In summary, a greater dose of ECP increased the percentage of animals expressing estrus, but it did not benefit the reproductive function of lactating dairy cows during the summer, regardless if animals were assigned to a TAI or timed-embryo transfer (TET) protocol.

The objective of this study was to compare the effects of different lengths of ovulation synchronization protocols using 2 controlled internal drug release (CIDR) devices on ovarian dynamics and pregnancy outcomes in lactating dairy cows. Lactating Holstein cows (n = 1,979) were randomly assigned to receive timed artificial insemination (TAI; d 0) following 1 of 2 treatments: (1) 9-d protocol (n = 988; 9D) with 2 intravaginal devices containing 1.9 g of progesterone (CIDR) and 2.0 mg of estradiol benzoate on day -11; 25 mg (i.m.) of dinoprost tromethamine (PG) and withdrawal of 1 CIDR on d -4; 1.0 mg (i.m.) of estradiol cypionate, second CIDR withdrawal, and PG on d -2; and TAI on d 0 and (2) 10-d protocol (n = 991; 10D) with 2 CIDR and 2.0 mg of estradiol benzoate on d -12; 25 mg of PG and withdrawal of 1 CIDR on d -4; 1.0 mg of estradiol, second CIDR withdrawal, and PG on d -2; and TAI on d 0. There was no effect of protocol on estrus detection, whereas a greater percentage of cows from 10D had ovulated close to TAI [no corpus luteum (CL) at AI and a CL at d 7] versus cows assigned to 9D protocol. A protocol × heat stress (average cow temperature ≥39.1°C on day of AI and d 7) interaction was observed in a manner that pregnancy per AI (P/AI) was greater in non-heat-stressed 10D versus 9D cows, whereas P/AI did not differ when cows were under heat stress. Furthermore, 10D protocol did not increase P/AI when all cows that received AI were included in the analysis or in cows that ovulated near TAI. However, animals assigned to 9D without any event of heat stress had a reduced P/AI when compared with cows assigned to 10D without heat stress. A protocol × CL presence at the beginning of the protocol interaction was observed and cows with a CL at the beginning of the protocol had a greater P/AI in 10D versus 9D; meanwhile, in cows without a CL, no differences on P/AI were observed. The protocol × CL presence at the beginning of the protocol interaction on P/AI was also observed for cows that ovulated near TAI. A greater percentage of cows assigned to 9D had follicles of medium size (13-15.9 mm), and greater percentage of cows assigned to 10D had larger follicles (>16 mm). Increasing the length of an estradiol with progesterone-based ovulation synchronization protocol (10D vs. 9D) increased the proportion of cows with larger follicles (>16 mm) and increased P/AI in cows without heat stress and in cows with a CL at beginning of the protocol. Moreover, the 10D protocol increased the proportion of cows with ovulation near TAI, demonstrating the effectiveness of this protocol in improving the reproductive performance of lactating Holstein cows.

The bovine appeasing substance (BAS) is expected to have calming effects in cattle experiencing stressful situations. Therefore, this study investigated the impacts of BAS administration during two of the most stressful events within beef production systems: weaning and feedlot entry. In experiment 1, 186 Bos indicus-influenced calves (73 heifers, 113 bulls) were weaned at 211 ± 1 days of age (day 0). At weaning, calves were ranked by sex and BW, and assigned to receive BAS (Nutricorp, Araras, SP, Brazil; n = 94) or water (CON; n = 92). Treatments (5 ml) were topically applied to the nuchal skin area of each animal. Calf BW was recorded and samples of blood and tail-switch hair were collected on days 0, 15 and 45. Calves that received BAS had greater (P < 0.01) BW gain from day 0 to 15 compared with CON. Overall BW gain (days 0 to 45) and BW on days 15 and 45 were also greater (P ≤ 0.03) in BAS v. CON. Plasma haptoglobin concentration was less (P < 0.01) in BAS v. CON on day 15, whereas cortisol concentrations in plasma and tail-switch hair did not differ between treatments (P ≥ 0.13). In experiment 2, 140 B. indicus-influenced bulls (∼27 months of age) from 2 different pasture-based systems (70 bulls/origin) were transported to a commercial feedlot (≤ 200-km transport; day -1). On day 0, bulls were ranked by source and BW, and assigned to receive BAS (n = 70) or CON (n = 70) and the same sampling procedures as in experiment 1. Bulls receiving BAS had greater (P = 0.04) BW gain from day 0 to 15, but less (P < 0.01) BW gain from day 15 to 45 compared to CON. No other treatment effects were detected (P > 0.14). Therefore, BAS administration to beef calves alleviated the haptoglobin response associated with weaning, and improved calf growth during the subsequent 45 days. Administration of BAS to beef bulls at feedlot entry improved BW gain during the initial 15 days, but these benefits were not sustained throughout the 45-day experiment.

The objectives of this study were to determine if the utilization of a presynchronization strategy would improve fertility at first artificial insemination (AI) during an E2/P4 ovulation synchronization protocol with or without GnRH administration at the beginning of the protocol. This experiment was conducted using cows (n = 665) at their first postpartum service and the following breeding treatment: CIDR insertion and 2.0 mg of estradiol benzoate (EB) on day -11; 25 mg dinoprost tromethamine (PG) on day -4; PG, CIDR withdrawal, and 1.0 mg of estradiol cypionate (ECP) on day -2; timed-AI on day 0. At 31 ± 3 days postpartum, cows were randomly allocated to one of three treatments on a weekly basis: 1) -P + GnRH: cows assigned to the breeding protocol with 100 µg of GnRH on day -11, 2) P + GnRH: cows assigned to a presynchronization protocol using CIDR insertion +2 mg EB on day -28, PG + ECP and CIDR withdrawal on day -21, and beginning of the breeding protocol plus GnRH (100 µg) on day -11, and 3) +P-GnRH: cows assigned to a presynchronization protocol and the breeding treatment without GnRH on day -11. No treatment effects were observed on P/AI at the pregnancy diagnoses on days 32 and 60, or for pregnancy losses between days 32 and 60 of pregnancy whether analyses included all cows or only cows that ovulated near TAI. Moreover, milk yield negatively affected P/AI. Cows with greater circulating P4 concentrations on day -4 had greater P/AI on day 60. Cows without CL on day -11 had a reduced P/AI and this effect was more significant in cows not treated with GnRH. Cows assigned to -P + GnRH had the lowest circulating P4 concentration on day -4 (3.4 ± 0.16 ng/mL), followed by + P-GnRH (4.56 ± 0.17 ng/mL), and +P + GnRH (5.08 ± 0.17 ng/mL) cohorts. The data of the current study suggest that the combination of a Presynch and GnRH administration at the beginning of a TAI protocol was the most effective way to increase the % of cows with a functional CL and with elevated circulating P4 concentrations at the time of PG treatment.

Excitable temperament disrupts physiological events required for reproductive development in cattle, but no research has investigated the impacts of temperament on growth and puberty attainment in Bos indicus females. Hence, this experiment evaluated the effects of temperament on growth, plasma cortisol concentrations and puberty attainment in B. indicus heifers. A total of 170 Nelore heifers, weaned 4 months before the beginning of this experiment (days 0 to 91), were managed in two groups of 82 and 88 heifers each (mean ± SE; initial BW=238±2 kg, initial age=369±1 days across groups). Heifer temperament was evaluated via exit velocity on day 0. Individual exit score was calculated within each group by dividing exit velocity into quintiles and assigning heifers with a score from 1 to 5 (1=slowest; 5=fastest heifer). Heifers were classified according to exit score as adequate (ADQ, n=96; exit score⩽3) or excitable temperament (EXC, n=74; exit score>3). Heifer BW, body condition score (BCS) and blood samples were obtained on days 0, 31, 60 and 91. Heifer exit velocity and score were recorded again on days 31, 60 and 91. Ovarian transrectal ultrasonography was performed on days 0 and 10, 31 and 41, 60 and 70, 81 and 91 for puberty evaluation. Heifer was declared pubertal at the first 10-day interval in which a corpus luteum was detected. Exit velocity and exit score obtained on day 0 were correlated (r⩾0.64, P<0.01) with evaluations on days 31, 60 and 91. During the experiment, ADQ had greater (P<0.01) mean BCS and BW gain, and less (P<0.01) mean plasma cortisol concentration compared with EXC heifers. Temperament × time interactions were detected (P<0.01) for exit velocity and exit score, which were always greater (P<0.01) in EXC v. ADQ heifers. A temperament × time interaction was also detected (P=0.03) for puberty attainment, which was delayed in EXC v. ADQ heifers. At the end of the experiment, a greater (P<0.01) proportion of ADQ were pubertal compared with EXC heifers. In summary, B. indicus heifers classified as EXC had reduced growth, increased plasma cortisol concentrations and hindered puberty attainment compared to ADQ heifers. Moreover, exit velocity may serve as temperament selection criteria to optimize development of B. indicus replacement heifers.

Eighty-four multiparous, nonlactating, pregnant Angus × Hereford cows were ranked by pregnancy type (56 AI and 28 natural service), BW, and BCS and allocated to 21 drylot pens at the end of their second trimester of gestation (d 0). Pens were assigned to receive forage-based diets containing 1) sulfate sources of Cu, Co, Mn, and Zn (INR); 2) an organic complexed source of Cu, Mn, Co, and Zn (AAC; Availa 4; Zinpro Corporation, Eden Prairie, MN); or 3) no supplemental Cu, Co, Mn, and Zn (CON). Diets were offered from d 0 until calving and formulated to meet requirements for energy, protein, macrominerals, Se, I, and vitamins. The INR and AAC diets provided the same daily amount of Cu, Co, Mn, and Zn. Cow BW and BCS were recorded and liver samples were collected on d -10 and 2 wk (d 75) before the calving season. Within 3 h after calving, calf BW was recorded, liver samples were collected, and the expelled placenta was retrieved ( = 47 placentas). Calves were weaned on d 283 of the experiment, preconditioned for 45 d (d 283 to 328), transferred to a growing lot on d 328, and moved to a finishing lot on d 440 where they remained until slaughter. Liver Co, Cu, and Zn concentrations on d 75 were greater ( ≤ 0.05) for INR and AAC cows compared with CON cows, whereas INR cows had reduced ( = 0.04) liver Co but greater ( = 0.03) liver Cu compared with AAC cows. In placental cotyledons, Co concentrations were greater ( ≤ 0.05) in AAC and INR cows compared with CON cows, whereas Cu concentrations were increased ( = 0.05) only in AAC cows compared with CON cows. Calves from INR and AAC cows had greater ( < 0.01) liver Co concentrations at birth compared with calves from CON cows. Liver Cu and Zn concentrations at birth were greater ( ≤ 0.05) in calves from AAC cows compared with cohorts from CON cows. Weaning BW was greater ( ≤ 0.05) in calves from AAC cows compared with cohorts from CON cows, and this difference was maintained until slaughter. In the growing lot, calves from AAC cows had reduced ( < 0.01) incidence of bovine respiratory disease compared with CON and INR cohorts. Collectively, these results suggest that feeding the AAC diet to late-gestating beef cows stimulated programming effects on postnatal offspring growth and health compared with the CON diet. Therefore, supplementing late-gestating beef cows with an organic complexed source of Co, Cu, Zn, and Mn instead of no supplementation appears to optimize offspring productivity in beef production systems.

The aim of this study was to test the effect of expression of estrus at artificial insemination (AI) on endometrium, conceptus, and CL gene expression of beef cows. Thirty-six multiparous nonlactating Nelore cows were enrolled on an estradiol- and progesterone (P4)-based timed AI protocol (AI = Day 0) and then slaughtered for the endometrium, CL, and conceptus collection on Day 19. The animals were retrospectively grouped on the basis of cows that (1) showed signs of estrus near AI (n = 19; estrus) and (2) did not show any signs of estrus (n = 17; nonestrus). Body condition score, blood sampling, and ultrasound examination were performed on Days 0, 7, and 18 of the experiment followed by messenger RNA extraction and quantitative reverse transcription polymerase chain reaction analysis of 58 target genes. Data were checked for normality and analyzed by ANOVA for repeated measures using proc GLM, MIXED, and UNIVARIATE of SAS. Only pregnant cows were included in the analyses (n = 12; nonestrus, n = 11). Estrous expression had no correlation with parameters such as body condition score, preovulatory follicle and CL diameter, P4 concentration in plasma on Days 7 and 18 after AI, and interferon-tau concentration in the uterine flushing (P > 0.15); however, a significant increase was observed in conceptus size from cows that expressed estrus (P = 0.02; 38.3 ± 2.8 vs. 28.2 ± 2.9 mm). The majority of transcripts affected by estrous expression in the endometrium belong to the immune system and adhesion molecule family (MX1, MX2, MYL12A, MMP19, CXCL10, IGLL1, and SLPI; P ≤ 0.05), as well as those related with prostaglandin synthesis (OTR and COX-2; P ≤ 0.05). Genes related to apoptosis, P4 synthesis, and prostaglandin receptor were downregulated (CYP11A, BAX, and FPr; P < 0.05) in the CL tissue of cows that expressed estrus. In addition, four genes were identified as differentially expressed in the 19-day-old conceptus from cows that expressed estrus (ISG15, PLAU, BMP15, and EEF1A1; P < 0.05). There was also a significant effect of Day 7 concentration of P4 mainly affecting the immune system, adhesion molecules, and wnt signaling pathway of the endometrium (IGLL1, MX2, SLPI, TRD, APC, WNT2, GLYCAM1, and MYL12A; P < 0.05). A significant interaction between estrous expression and P4 concentration on Day 7 was more pronounced in immune system genes (MX1, MX2, TRD, SLPI, and IGLL1; P < 0.05). This study reported that estrous expression at the time of AI favorably altered the gene expression profile in reproductive tissues during the preimplantation phase toward a more receptive state to the elongating conceptus. These effects seem to be more evident in the endometrium during the time of dynamic remodeling for embryo implantation.

The objective of this study was to evaluate intake, metabolic, inflammatory, and acute-phase responses in beef heifers vaccinated against pathogens that cause bovine respiratory disease (BRD). Eighteen weaned Angus heifers (initial BW 257 ± 3 kg; initial age 245 ± 2 d) were ranked by BW and allocated to 2 groups, which were assigned to 2 experiments of 7 d and the following treatments on d 1 of each experiment: 1) revaccinated against infectious bovine rhinotracheitis virus, parainfluenza-3 virus, bovine respiratory syncytial virus, bovine viral diarrhea Types 1 and 2 viruses, and (VAC; 2 mL [s.c.]) and 2) receiving a 2-mL s.c. injection of 0.9% sterile saline (CON). The group receiving VAC in Exp. 1 was assigned to CON in Exp. 2 and vice versa. Heifers were weaned 21 d before Exp. 1, when they all received the first dose of the aforementioned vaccine. Heifers were maintained in individual pens and offered free-choice mixed alfalfa-grass hay and 3.5 kg/d (DM basis) of a corn-based supplement throughout the study. During Exp. 1, hay and concentrate intake were evaluated daily. During Exp. 2, blood samples were collected before (-2 and 0 h) and at 2, 4, 6, 8, 12, 16, 24, 36, 48, 60, 72, 96, 120, 144, and 168 h after treatment administration. In Exp. 1, treatment × day interactions were detected ( < 0.01) for forage intake and total DMI; these parameters were reduced ( ≤ 0.05) in VAC heifers compared with CON heifers on d 1 and 2 by an average of 1.7 and 0.8 kg (DM basis), respectively. In Exp. 2, mean serum tumor necrosis factor α (TNFα) concentration was greater ( = 0.05) in VAC heifers compared with CON heifers and treatment × hour interactions were detected for all plasma variables ( ≤ 0.02), whereas a similar tendency was detected ( = 0.09) for blood α mRNA expression. Haptoglobin concentrations were greater ( ≤ 0.05) in VAC heifers compared with CON heifers from 16 to 120 h. Blood α mRNA expression was greater ( = 0.05) in VAC heifers compared with CON heifers at 12 h. Cortisol concentrations were greater ( ≤ 0.05) in VAC heifers compared with CON heifers from 2 to 16 h. Insulin concentration was greater ( = 0.02) in VAC heifers compared with CON heifers at 2 h. Leptin concentrations were greater ( ≤ 0.05) in VAC heifers compared with CON heifers from 6 to 16 h. In conclusion, vaccinating beef heifers against BRD pathogens decreased forage intake and total DMI during the 2 d following vaccination in Exp. 1, which can be associated with transient metabolic, inflammatory, and acute-phase responses elicited by vaccination in Exp. 2.

This experiment compared growth, physiological, and reproductive responses of beef heifers with (MI) or without (CON) access to a creep-feeder, as a manner to stimulate metabolic imprinting while nursing their dams. On day 0, 60 Angus × Hereford heifers were ranked by BW and age (140 ± 3 kg and 68±3 days), and assigned to pairs so all ranking criteria were similar between heifers within each pair. On day 1, pairs were randomly assigned to MI (n=15) or CON (n=15). From day 1 to 51, MI pairs and their dams were allocated to 15 drylot pens where heifers had ad libitum access to a corn-based supplement through a creep-feeder. The CON pairs and their dams were maintained in an adjacent single drylot pen. From day 52 to 111, treatments were managed as a single group on a semiarid range pasture. On day 111, heifers were weaned and allocated to two pastures (one pasture/treatment), receiving hay and a corn-based concentrate until day 326. Heifer BW was recorded before and at the end of the creep-feeding period (day 1 to 51), and on days 112 and 326. On days 0, 51, 111, 187, 261, and 325, jugular blood was collected and real-time ultrasonography for longissimus muscle depth and backfat thickness assessment was performed. Blood was also collected every 10 days from days 113 to 323 for puberty evaluation via plasma progesterone. Liver and subcutaneous fat biopsies were performed on days 51, 111, 261 and 325. Average daily gain was greater (P<0.01) for MI than CON from day 1 to 51, tended (P=0.09) to be greater for CON than MI from day 112 to 326, while BW on day 326 was similar between treatments. On day 51, MI had greater (P ⩽ 0.01) plasma IGF-I and glucose concentrations, as well as mRNA expression of hepatic pyruvate carboxylase and adipose fatty acid synthase than CON. On days 261 and 325, plasma insulin concentrations were greater (P ⩽ 0.03) in CON than MI. Mean mRNA expression of hepatic IGF-I and adipose peroxisome proliferator-activated receptor gamma were greater (P ⩽ 0.05) in MI than CON. No treatment effects were detected for puberty attainment rate. In conclusion, supplementing nursing heifers via creep-feeding for 50 days altered physiological and biochemical variables suggestive of a metabolic imprinting effect, but did not hasten their puberty attainment.

The objective of this experiment was to determine if frequency of protein supplementation impacts physiological responses associated with reproduction in beef cows. Fourteen nonpregnant, nonlactating beef cows were ranked by age and BW and allocated to 3 groups. Groups were assigned to a 3 × 3 Latin square design, containing 3 periods of 21 d and the following treatments: 1) soybean meal supplementation daily (D), 2) soybean meal supplementation 3 times/week (3WK), and 3) soybean meal supplementation once/week (1WK). Within each period, cows were assigned to an estrus synchronization protocol: 100 µg of GnRH + controlled internal drug release device (CIDR) containing 1.38 g of progesterone (P4) on d 1, 25 mg of PGF2α on d 8, and CIDR removal + 100 µg of GnRH on d 11. Grass-seed straw was offered for ad libitum consumption. Soybean meal was individually supplemented at a daily rate of 1 kg/cow (as-fed basis). Moreover, 3WK was supplemented on d 0, 2, 4, 7, 9, 11, 14, 16, and 18 whereas 1WK was supplemented on d 4, 11, and 18. Blood samples were collected from 0 (before) to 72 h after supplementation on d 11 and 18 and analyzed for plasma urea-N (PUN). Samples collected from 0 to 12 h were also analyzed for plasma glucose, insulin, and P4 (d 18 only). Uterine flushing fluid was collected concurrently with blood sampling at 28 h for pH evaluation. Liver biopsies were performed concurrently with blood sampling at 0, 4, and 28 h and analyzed for mRNA expression of carbamoyl phosphate synthetase I (CPS-I; h 28) and CYP2C19 and CYP3A4 (h 0 and 4 on d 18). Plasma urea-N concentrations were greater (P < 0.01) for 1WK vs. 3WK from 20 to 72 h and greater (P < 0.01) for 1WK vs. D from 16 to 48 h and at 72 h after supplementation (treatment × hour interaction, P < 0.01). Moreover, PUN concentrations peaked at 28 h after supplementation for 3WK and 1WK (P < 0.01) and were greater (P < 0.01) at this time for 1WK vs. 3WK and D and for 3WK vs. D. Expression of CPS-I was greater (P < 0.01) for 1WK vs. D and 3WK. Uterine flushing pH tended (P ≤ 0.10) to be greater for 1WK vs. 3WK and D. No treatment effects were detected (P ≥ 0.15) on expression of CYP2C19 and CYP3A4, plasma glucose, and P4 concentrations, whereas plasma insulin concentrations were greater (P ≤ 0.03) in D and 3WK vs. 1WK. Hence, decreasing frequency of protein supplementation did not reduce uterine flushing pH or plasma P4 concentrations, which are known to impact reproduction in beef cows.

This experiment evaluated the effects of meloxicam administration on physiological and performance responses of transported cattle during feedlot receiving. Eighty-four Angus × Hereford steers were ranked by BW on d -10 and assigned to 21 dry lot pens. From d -10 to 0, pens were fed alfalfa-grass hay ad libitum and 2.4 kg/steer daily (DM basis) of a corn-based concentrate. On d 0, pens were randomly assigned to 1) transport for 1,440 km in a livestock trailer and oral administration of meloxicam (1 mg/kg of BW) at loading (d 0), unloading (d 1), and daily from d 2 to 7 of feedlot receiving (MEL; n = 7); 2) the same transportation and treatment schedule of MEL but oral administration of lactose monohydrate (1 mg/kg of BW) instead of meloxicam (TRANS; n = 7); or 3) no transport and oral administration of lactose monohydrate (1 mg/kg of BW) concurrently with treatment administration to MEL and TRANS (CON; n = 7). Upon arrival (d 1), MEL and TRANS steers returned to their pens for a 21-d feedlot receiving with the same diet offered from d -10 to 0. Treatments were administered to steers via oral drench on d 0 and 1 or mixed daily with the concentrate from d 2 to 7. Full BW was recorded before (d -2, -1, and 0) treatment application and at the end of experiment (d 20, 21, and 22) for ADG calculation. Daily DMI was recorded from d 1 to 21. Blood samples were collected on d 0, 1, 3, 5, 7, 10, 14, and 21. During the initial 7 d of feedlot receiving, hay and total DMI were reduced (P ≤ 0.03) in TRANS vs. CON and MEL and similar between CON and MEL (P ≥ 0.26), whereas concentrate DMI did not differ (P = 0.16) among treatments. Mean ADG was reduced (P ≤ 0.03) in TRANS vs. MEL and CON but similar (P = 0.82) between MEL and CON. Moreover, TRANS had reduced G:F vs. CON (P = 0.01) and MEL (P = 0.05), whereas G:F was similar (P = 0.39) between CON and MEL. Serum NEFA concentrations were greater (P < 0.01) for TRANS and MEL vs. CON on d 1. Plasma haptoglobin concentrations were greater (P ≤ 0.03) for TRANS vs. CON and MEL on d 5 and greater (P ≤ 0.03) for CON vs. TRANS on d 10. Plasma ceruloplasmin concentrations were greater (P ≤ 0.04) for TRANS vs. CON on d 3, 5, 7, 10, and 14, greater (P ≤ 0.03) for TRANS vs. MEL on d 5 and 7, and also greater (P = 0.05) for MEL vs. CON on d 3. In conclusion, meloxicam administration to feeder steers modulated the haptoglobin and ceruloplasmin responses and prevented the performance losses caused by long-distance transportation.

Two experiments evaluated the influence of supplement composition on ruminal forage disappearance, performance, and physiological responses of Angus × Hereford cattle consuming a low-quality cool-season forage (8.7% CP and 57% TDN). In Exp. 1, 6 rumen-fistulated steers housed in individual pens were assigned to an incomplete 3 × 2 Latin square design containing 2 periods of 11 d each and the following treatments: 1) supplementation with soybean meal (PROT), 2) supplementation with a mixture of cracked corn, soybean meal, and urea (68:22:10 ratio, DM basis; ENER), or 3) no supplementation (CON). Steers were offered meadow foxtail (Alopecurus pratensis L.) hay for ad libitum consumption. Treatments were provided daily at 0.50 and 0.54% of shrunk BW/steer for PROT and ENER, respectively, to ensure that PROT and ENER intakes were isocaloric and isonitrogenous. No treatment effects were detected on rumen disappearance parameters of forage DM (P ≥ 0.33) and NDF (P ≥ 0.66). In Exp. 2, 35 pregnant heifers were ranked by initial BW on d -7 of the study, allocated into 12 feedlot pens (4 pens/treatment), and assigned to the same treatments and forage intake regimen as in Exp. 1 for 19 d. Treatments were fed once daily at 1.77 and 1.92 kg of DM/heifer for PROT and ENER, respectively, to achieve the same treatment intake as percent of initial BW used in Exp. 1 (0.50 and 0.54% for PROT and ENER, respectively). No treatment effects (P = 0.17) were detected on forage DMI. Total DMI was greater (P < 0.01) for PROT and ENER compared with CON and similar between PROT and ENER (P = 0.36). Accordingly, ADG was greater (P = 0.01) for PROT compared with CON, tended to be greater for ENER compared with CON (P = 0.08), and was similar between ENER and PROT (P = 0.28). Heifers receiving PROT and ENER had greater mean concentrations of plasma glucose (P = 0.03), insulin (P ≤ 0.09), IGF-I (P ≤ 0.04), and progesterone (P = 0.01) compared to CON, whereas ENER and PROT had similar concentrations of these variables (P ≥ 0.15). A treatment × hour interaction was detected (P < 0.01) for plasma urea N (PUN), given that PUN concentrations increased after supplementation for ENER and PROT (time effect, P < 0.01) but did not change for CON (time effect, P = 0.62). In conclusion, beef cattle consuming low-quality cool-season forages had similar ruminal forage disappearance and intake, performance, and physiological status if offered supplements based on soybean meal or corn at 0.5% of BW.

Heifers were offered meadow foxtail (Alopecurus pratensis L.) hay for ad libitum consumption during the experiment (d -10 to 160). Beginning on d 0, PROT and ENER were provided daily at a rate of 1.30 and 1.40 kg of DM/heifer to ensure that PROT and ENER intakes were isocaloric and isonitrogenous. Hay and total DMI were recorded for 5 consecutive days during each month of the experiment. Blood was collected every 10 d for analysis of plasma progesterone to evaluate puberty attainment. Blood samples collected on d -10, 60, 120, and 150 were also analyzed for plasma concentrations of plasma urea N (PUN), glucose, insulin, IGF-I, NEFA, and leptin. Liver samples were collected on d 100 from 2 heifers/pen and analyzed for mRNA expression of genes associated with nutritional metabolism. No treatment effect was detected (P = 0.33) on forage DMI. Total DMI, ADG, and mean concentrations of glucose, insulin, and IGF-I as well as hepatic mRNA expression of IGF-I and IGFBP-3 were greater (P ≤ 0.02) for PROT and ENER compared with CON and similar between PROT and ENER (P ≥ 0.13). Mean PUN concentrations were also greater (P < 0.01) for PROT and ENER compared with CON, whereas PROT heifers had greater (P < 0.01) PUN compared with ENER. Plasma leptin concentrations were similar between ENER and PROT (P ≥ 0.19) and greater (P ≤ 0.03) for ENER and PROT compared with CON on d 120 and 150 (treatment × day interaction, P = 0.03). Hepatic mRNA expression of mitochondrial phosphoenolpyruvate carboxykinase was greater (P = 0.05) in PROT compared with CON and ENER and similar between CON and ENER (P = 0.98). The proportion of heifers pubertal on d 160 was greater (P < 0.01) in ENER compared with PROT and CON and similar between PROT and CON (P = 0.38). In conclusion, beef heifers consuming a low-quality cool-season forage had a similar increase in DMI, growth, and overall metabolic status if offered supplements based on soybean meal or corn at 0.5% of BW.

RESUMO

The objective of this experiment was to compare fatty acid (FA) concentrations in plasma and reproductive tissues as well as hormones and expression of genes associated with pregnancy establishment in beef cows supplemented or not with Ca salts of soybean oil (CSSO) beginning after timed AI. Ninety nonlactating multiparous Nelore (Bos indicus) cows were timed inseminated on d 0 of the experiment and divided into 18 groups of 5 cows/group. Groups were randomly assigned to receive (as-fed basis) 100 g of a protein-mineral mix plus 100 g of ground corn per cow daily in addition to 1) 100 g/cow daily of CSSO (n = 9) or 2) 100 g/cow daily of kaolin (CON; rumen-inert indigestible substance; n = 9). All groups were maintained in a single Brachiaria brizanta pasture (24 ha) with ad libitum access to forage and water. However, groups were segregated daily and offered treatments individually at the working facility during the experimental period (d 0 to 18). Blood samples were collected and transrectal ultrasonography was performed to verify ovulation and estimate corpus luteum (CL) volume immediately before AI (d 0) and on d 7 and 18 of the experiment. On d 19, 36 cows (18 cows/treatment; 2 cows/group) diagnosed without the presence of a CL on d 0 but with a CL greater than 0.38 cm(3) in volume on d 7 and 18 were slaughtered for collection of conceptus, uterine luminal flushing, and tissue samples from the CL and endometrium. Cows receiving CSSO had greater concentrations of linoleic and other ω-6 FA in plasma (P < 0.01), endometrium (P ≤ 0.05), CL (P ≤ 0.05), and conceptus (P ≤ 0.08) compared to CON. On d 7 of the experiment, CSSO-supplemented cows had greater plasma progesterone concentrations (P < 0.01) and CL volume (P = 0.02) compared to CON, whereas no treatment effects were detected (P ≥ 0.15) for these parameters on d 18 (treatment × day interaction; P < 0.01). Cows receiving CSSO tended (P = 0.09) to have greater concentrations of interferon-tau in the uterine flushing media compared with CON. However, no treatment effects were detected for mRNA expression genes associated with pregnancy establishment in endometrial, CL, and conceptus samples (P ≥ 0.12). In summary, supplementing beef cows with 100 g of CSSO beginning after AI favored incorporation of ω-6 FA into their circulation, reproductive tissues, and conceptus, without impacting expression of genes associated with pregnancy establishment on d 19 of gestation.

RESUMO

This experiment evaluated temperament, vaginal temperature, and plasma cortisol in beef cows from wolf-naïve and wolf-experienced origins that were subjected to a simulated wolf encounter. Multiparous, pregnant, nonlactating Angus-crossbreed cows from the Eastern Oregon Agricultural Research Center located near Burns, OR (CON; n = 50), and from a commercial operation near Council, ID (WLF; n = 50), were used. To date, grey wolves are not present around Burns, OR, and thus CON were naïve to wolves. Conversely, wolves are present around Council, ID, and WLF cows were selected from a herd that had experienced multiple confirmed wolf-predation episodes from 2008 to 2012. Following a 50-d commingling and adaptation period, CON and WLF cows were ranked by temperament, BW, and BCS and allocated to 5 groups (d 0; 10 CON and 10 WLF cows/group). Groups were individually subjected to the experimental procedures on d 2 (n = 3) and d 3 (n = 2). Before the simulated wolf encounter, cow temperament was assessed and blood samples and vaginal temperatures (using intravaginal data loggers) were collected (presimulation assessments). Cows were then sorted by origin, moved to 2 adjacent drylot pens (10 WLF and 10 CON cows/pen), and subjected to a simulated wolf encounter event for 20 min, which consisted of 1) cotton plugs saturated with wolf urine attached to the drylot fence, 2) continuous reproduction of wolf howls, and 3) 3 leashed dogs that were walked along the fence perimeter. Thereafter, WLF and CON cows were commingled and returned to the handling facility for postsimulation assessments, which were conducted immediately after exposure to wolf-urine-saturated cotton plugs, wolf howl reproduction, and 20-s exposure to the 3 dogs while being restrained in a squeeze chute. Chute score, temperament score, and plasma cortisol concentration increased (P ≤ 0.01) from pre- to postsimulation assessment in WLF but did not change in CON cows (P ≥ 0.19). Exit velocity decreased (P = 0.01) from pre- to postsimulation assessment in CON but did not change (P = 0.79) in WLF cows. In addition, WLF cows had a greater (P = 0.03) increase in temperature from pre- to postsimulation assessments compared with CON cows. In conclusion, the simulated wolf encounter increased excitability and fear-related physiological stress responses in cows that originated from a wolf-experienced herd but not in cows that originated from a wolf-naïve herd.
